High Sensitivity Lc-ms/ms Method For Pthrp Analysis

ID U-5729

Category Diagnostics

Subcategory Biomarkers

Brief Summary

A revolutionary LC-MS/MS method offering high sensitivity and specificity for measuring parathyroid hormone-related peptide (PTHrP) in biological samples

Problem Statement

Currently available methods for analysis of PTHrP, including Radioimmunoassay (RIA) and ELISA, have poor analytical performance, underestimate PTHrP concentration in patient samples, and have inadequate clinical sensitivity and specificity, leading to false negatives.

Technology Description

This technology introduces a method for measuring parathyroid hormone-related peptide (PTHrP) in biological samples, utilizing LC-MS/MS along with an isotope-labeled internal standard. By extracting PTHrP peptides, digesting them proteolytically, and analyzing specific digestion products, this method achieves unprecedented sensitivity and specificity in PTHrP quantification.
• Diagnostic testing for hypercalcemia and related conditions, improving the detection of PTHrP-secreting malignancies.
• Research tool for studying the role of PTHrP in calcium regulation, bone development, and other physiological functions.
• Potential use in drug development and therapeutic monitoring, particularly for treatments affecting calcium homeostasis.

Stage of Development

Clinical Validation

Benefit

  • High sensitivity capable of detecting PTHrP concentrations as low as 0.5 pmol/L or less.
  • High specificity through the use of isotope-labeled internal standards for accurate quantification.
  • Rapid sample preparation process, completed in 2 hours or less to minimize PTHrP degradation.
  • Utilization of affinity extraction for selective PTHrP peptide isolation, enhancing measurement precision.
  • Capability to process without the need for reduction or alkylation reactions, simplifying the preparation steps.
